概要: Incidence and mortality rates of renal cell carcinoma (RCC) in Japan are increasing and approximately 3000 person died of RCC. Radical surgical removal of small RCC, frequently denoted as less than 4 cm, leads to favorable prognosis. On the contrary, virtually all patients with metastasis succumb to the disease partly because RCC is refractory to chemotherapy or irradiation. RCC has unique character including rather frequent spontaneous regression cases reported, rather higher response rates to administration of cytokines. These indicate needs for brand new treatment strategy based on the individual properties of each RCC. 続きを見る

概要: Propionibacterium acnes has been implicated as the etiologic agent of sarcoidosis since the isolation of this bacterium at a high incidence from sarcoidosis specimens in 1978. However, this possibility has not been established or accepted widely yet. We have obtained the P. acnes strains isolated in that study. Based on the idea that inflammatory cytokines are involved in granuloma formation characteristic for pathology of sarcoidosis, we have compared the cytokine-inducing ability among various strains. [Method] Peripheral blood mononuclear cells were obtained from sarcoidosis patients and healthy controls. Cells were stimulated with P. acnes strains in vitro, and inflammatory cytokines were determined by using cytokine-specific EIA. The effect of T-cell depletion was examined. [Result] Th1-type of antigen-specific response was not observed when patient cells were stimulated with P. acnes. Strains from sarcoidosis specimen showed a significantly higher activity for induction of IL-12 and IFN-γ for whole mononuclear cells and also for T cell-depleted population. [Conclusion] The results suggested that particular P. acnes strains capable of inducing a high level of inflammatory cytokine are involved in the pathogenesis of sarcoidosis. It was postulated that P. acnes first stimulate macrophages to produce IL-12 then in turn stimulate NK cells to produce IFN-γ which may contribute to granuloma formation. 続きを見る

概要: It was previously demonstrated that preoperative administration of anti-αβ T cell receptor monoclonal antibody (R73) ind uced specific unresponsiveness to heart and kidney allografts in a fully MHC incompatible rat strain. In this study, the part of this mechanism was examined by molecular technique. Heterotopic heart and orthtopic left lung transplantation were performed. Experimental groups were divided as follows:Group 1, control group which were injected control antibody once a day for two consecutive days, beginning two days before transplantation. Group 2, preoperative treatment group which were injected R73 for two days as group 1. Group 3, postoperative treatment group which were injected R73 once a day for seven consecutive days, beginning on the day of transplantation. Heart grafts survived more than 200 days only in group 2. On the other hand, lung grafts were rejected with five days delay compared with control group. Phenotypic and functional analyses of T cells obtained from rats treated with R73 suggest that TCR molecules and function of T cells are transiently down-regulated at the time of transplantation. In RNase protection assay, expression of cytokine mRNA on grafts was suppressed only in group 2. These data suggest that exposure to alloantigen during T cell modulation suppress cytokine production which initiate and enhance graft rejection. In normal lung but not heart, cytokine and adhesion molecule were detected in mRNA level. This may be one of the reason of early graft rejection in lung transplantation. 続きを見る

概要: CD8-positive lymphocytes (CD8^+lym) and ICAM-1/LFA-1 interaction have been implicated in the glomerular accumulation of monocytes/macrophages (Mo/Mφ) and crescent formation in anti-glomerular basement membrane (GBM) glomerulonephritis of WKY rats. In the present study, the participation of CD8^+lym and inflammatory cytokines in the expression of ICAM-1 and the accumulation of Mo/Mφ in the glomeruli was examined. ICAM-1 expression and Mo/Mφ accumulation were markedly enhanced in the glomeruli after anti-GBM antibody injection. By depleting the CD8^+lym in the circulation, the increased expression of ICAM-1 and glomerular accumulation of Mo/Mφ were suppressed, indicating a crucial role of CD8^+lym in the enhancement of ICAM-1 expression and the Mo/Mφ accumulation. Expression of mRNA for IL-1β, TNFα and IFNγ, known to enhance ICAM-1 expression, was induced in the glomeruli at a mRNA level in a similar profile to that of ICAM-1 expression. The CD8^+lym depletion also suppressed the induction of mRNA expression for these cytokines except for IL-1β. These results indicate CD8^+lym up-regulate the expression of ICAM-1 probably through induction of TNFα and IFNγ and play a role in the accumulate Mo/Mφ. 続きを見る

概要: We investigated what the possible mediators or conditions that accelerate the expression of cellular adheison molecules on cultured endothelial cells are. Examined factors were native low density lipoprotein (LDL), oxidized LDL, glycated LDL, LDL-autoantibody immune complexes, and hypoxia. Adhesion molecules were ELAM-1, ICAM-1, ICAM-2, and VCAM-1. The present study showed that oxidized LDL, glycated LDL, LDL-autoantibody complexes, and hypoxia were mediaters that caused the expression of ELAM-1.
