概要: The lateral posterior complex (LP) of the thalamus receives a variety of different inputs from cortical and subcortical areas. We have previously demonstrated four types of vesicle containing profiles in this complex, and have identified GABA-ergic terminals. In this study, we examined the features by the light and electron microscopy of the immunoreactivity with antibodies raised against aspartate (Asp), glutamate (Glu), GABA, choline acetyltransferase (ChAT) and substance P (SP) in the LP complex. The following observations were found:(1) Asp and Glu;immunolabeled neurons were medium sized. Immunolabeled terminals were medium sized containing round vesicles and making asymmetrical synaptic contacts with dendritic profiles. (2) GABA;immunolabeled neurons were small. Immunolabeled terminals were small, contained pleomorphic synaptic vesicles and forming symmetrical synaptic contacts with dendritic profiles. (3) ChAT;no ChAT immunolabeled neurons were found. Immunolabeled terminals were small, and contained round vesicles making en passant, symmetrical and asymmetrical synaptic contacts with dendritic profiles. 概要: Endocrine cells in 84 cases of non-neoplastic mucosa, 122 adenomas and 211carcinomas of the colon and rectum were histochemically and immunohistochemically studied on their incidence, distribution and types. Moreover, histogenesis of endocrine cells in adenomas and carcinomas is discussed. Argyrophil-positive cells were found in 100 of 122 adenomas (82.0%), and 107 of 211 carcinomas (50.7%), both showing a higher incidence than ever reported. Most of the cells were made up of serotonin cells. Glicentin/glucagon cells and neurotensin cells were small in population. Somatostatin cells and pancreatic polypeptide cells were rare in population. Endocrine cells in adenomas and carcinomas were similar, in both the type and relative population of cells, to those of the non-neoplastic mucosa, although the population of endocrine cells in adenomas and carcinomas was influenced by differentiation of neoplastic cells and structures. These findings may suggest that endocrine cells in adenomas an. d carcinomas of the colon and rectum show similarity in cell types and population to those of the non-neoplastic mucosa, and that such endocrine cells are derived from primitive neoplastic cells with capability of multidirectional differentiation. 続きを見る

概要: To investigate diagnostic value of carcinombryonic antigen (CEA) staining in sweat gland tumors, the normal skin and several kinds of epithelial skin tumors were immunohistochemically examined by the immunoperoxidase technique using polyclonal anti-CEA antibodies (PCEA) and monoclonal anti-CEA antibody (MCEA). In the normal skin tissue, the secretory cells, ductal cells, duct cuticle and luminal materials of either eccrine glands or apocrine glands showed PCEA-positive reactions. None of the other skin components showed positive reactions to the PCEA. A weak MCEA-positive reaction was seen only in the dermal and intraepidermal portions of the sweat gland ducts. In the skin tumors, PCEA positive reactions were found in all the cases of extramammary Paget's disease and sweat gland tumors. In all the cases of the squamous cell carcinoma and Bowen's disease, the keratotic parts showed PCEA-positive reactions. These reactions became very weak or negative, when stained with MCEA. Most of the PCEA-positive materials in the skin tissues may be different from the original CEA. Although PCEA is still thought to be important as an immunohistochemical marker for the diagnosis of sweat gland tumors, there may be a heterogeneity of PCEA-reacting substances among normal skin and tumor tissues. 続きを見る

概要: Experimental allergic encephalomyelitis (EAE) is a T-cell mediated autoimmune disease inducible by immunization with brain specific antigens such as myelin basic protein (MBP) in complete Freund's adjuvant (CFA). Using EAE as an experimental model for human autoimmune disorders, we show here several experimental approaches and the results obtained by these approaches. They include 1) immunohistochemical examination of the lesions of EAE, 2) double immunofluorescent staining of the tissue in order to identify the nature of cells of interest, 3) passive EAE which facilitutes the examination of developing lesions of EAE, and 4) radiation bone marrow chimeras which were prepared for the detection of bone marrow-derived cells in the central nervous system. We also introduced limiting dilution analysis of MBP-reactive T cells and thymus-grafted chimeras. 続きを見る
