概要: To investigate the relationship between striatal dopaminergic function and behavior, we simultaneously examined the intr astriatal kinetics of the dopamine antagonist sulpiride and cataleptic behavior using in vivo voltammetry. First, we confirmed that sulpiride, a D2 antagonist, could be measured in vivo with this technique. Sulpiride current intensity was measured when the potential was increased from 750 to 850 mV. Then sulpiride was administered into the striatum of freely moving rats. Sulpiride levels increased and remained high for more than 3h. Cataleptic behavior also continued for 3h. The duration of cataleptic behavior correlated well with that increase in sulpiride. Sulpiride kinetics was compared with that of dopamine and 1-methyl-1, 4-phenylpyridi-nium(MPP+). Dopamine and MPP+ were administered intrastriatally and were taken up into dopaminergic neurons within 1h after the end of injection. These findings suggest that sulpiride is not taken up or metabolized. 続きを見る

概要: The relationship between the visual cortex and superior colliculus (SC) is integral in the elaboration of visually-guided behaviors. Although numerous regions of visual cortex send projections to SC, the lateral suprasylvian (LS) cortex is known to be a primary source of direct visual cortical afferents to the deep laminae of SC, in which tectospinal neurons are located. Our recent experiments provide anatomical and physiological evidence to support the idea that, in addition to a direct influence on deep lamina neurons, LS may modulate tectospinal neurons indirectly via its projection to the striatum. 続きを見る

概要: The neostriatum, nucleus accumbens and basal nucleus of meynert (bnM) in Alzheimer's disease (AD), progressive supranuclear palsy (PSP) and parkinsonism-dementia complex of Guam (GuamPDC) were examined immunohistologically, ultrastructurally, quantitatively and topographically. Compaired to neurologically normal controls, the number of large neurons was reduced by approximately 65% in the caudate nucleus and putamen in these diseases and especially, in AD and GuamPDC, by more than 90% in the nucleus accumbens. The decreased number of large neurons in the neostriatum was significantly correlated to that in the bnM in AD and GuarnPDC, but not in PSP. The remaining large neurons in the neostriatum and nucleus accumbens of these three diseases contained neurofibrillary tangles (NFTs). The NFTs were composed varying amounts of 21-to 25-nm wide paired helical filaments admixed with straight tubules (STs) in AD and GuamPDC, and composed mainly of STs in PSP. Extremely severe loss of large neurons in the nucleus accumbens in AD and GuamPDC may be linked to marked degeneration of the limbic and ventral tegmental areas and nucleus dorsal raphe. 続きを見る

概要: Projections from the visual cortical areas to the striatum and superior colliculus are believed to play integral roles in visual attentive and orienting behaviors. In the cat, the direct corticotectal and indirect corticostriatal (via the substanita nigra) projections of the lateral suprasylvian cortex (LS) are thought to converge on neurons situated in the deep laminae of the superior colliculus and exert their influences via the outputs of the superior colliculus (Norita et.
