monocytes/macrophages (Mo/Mφ) and crescent formation in anti-glomerular basement membrane (GBM) glomerulonephritis of WKY rats. In the present study, the participation of CD8^+lym and inflammatory cytokines in the expression of ICAM-1 and the accumulation of Mo/Mφ in the glomeruli was examined. ICAM-1 expression and Mo/Mφ accumulation were markedly enhanced in the glomeruli after anti-GBM antibody injection. By depleting the CD8^+lym in the circulation, the increased expression of ICAM-1 and glomerular accumulation of Mo/Mφ were suppressed, indicating a crucial role of CD8^+lym in the enhancement of ICAM-1 expression and the Mo/Mφ accumulation. Expression of mRNA for IL-1β, TNFα and IFNγ, known to enhance ICAM-1 expression, was induced in the glomeruli at a mRNA level in a similar profile to that of ICAM-1 expression. The CD8^+lym depletion also suppressed the induction of mRNA expression for these cytokines except for IL-1β. These results indicate CD8^+lym up-regulate the expression of ICAM-1 probably through induction of TNFα and IFNγ and play a role in the accumulate Mo/Mφ.

Adhesion molecules were ELAM-1, ICAM-1, ICAM-2, and VCAM-1. The present study showed that oxidized LDL, glycated LDL, LDL-autoantibody complexes, and hypoxia were mediaters that caused the expression of ELAM-1. It was also found that TNFα played a key role in the sequence.
