rentiate functionally into one of two subsets, Th1 or Th2. Th1 cells generate IL-2 and IFNγ and support the cellular immune responses. On the other hand, Th2 cells generate IL-4, IL-5 and IL-10 and support the humoral immune responses. A unique population of T cells that carry intermediate levels of TCR exist in the liver and other immune organs. These T cells express IL-2Rβ-chain and receptors of the NK cell lineage. Many investigators call them intTCR cells or NKT cells. Recently, it was demonstrated that NKT cells produce IL-4 and IFN-γ and regulate the differentiation of CD4^+ helper T cells into Th1 or Th2 cells. These findings indicate that NKT cells play an important role of immune regulation by cytokines.

were modulated during melanoma development in melanoma transgenic mice. Melanoma appeared in various organ of the transgenic mice at 15 wk of age. Extrathymic T cells with TCR of intermediate intensity increased in the liver, as the tumors advanced. Such cells infiltated the tumor lesions. In consequence of this study, it was proved that extrathymic T cells in the liver played important roles in tumor immunity.

tivity is the capacity to induce the IL-2 production of T cells. Interleukin 2 (IL-2), produced by activated T cells, induces the proliferation and differentiation on T and B cells. In this report, the signal requirement for the activation of T cells in C57BL/6 mice was examined in in vitro responses to mutant MHC class I antigen (H-2K^<bm1>). From our observations, activation of T cells responding to MHC class I antigen could be explained as follows. Lyt 2^+ T cells recognize directly the mutant MHC class I antigen and then IL-1 receptor (IL-1R) and IL-2 receptor (IL-2R) appear on the T cells. Accessory cells secrete IL-1 upon the binding of T cells. The secreted IL-1 may bind IL-1R on the responding T cells and induce production of IL-2, which binds to IL-2R. Thus, T cell proliferation and CTL (cytotoxic T lymphocyte) generation will proceed autonomously.
