概要: A specific chromosomal abnormality, the Philadelphia chromosome (Ph^1), is found in 90 to 95% of CML patients. Breakpoints on chromosome 22 in CML patients occur in a small region designated as the breakpoint cluster region (bcr). Using 3' bcr probe (a commerciall available bcr probe), we screened bcr rearrangements in patients with CMPD including two patients with Ph1 (+) ALL. The results showed that bcr rearrangements appear only in patients with CML, indicating that screening an abnormal bcr rearrangement is a useful tool to diagnose CML. In addision we analyzed the DNA from Ph1 (+) CML patients in blastic crisis to determine whether the differences in location of molecular of breakpoints within the bcr are assosiated with heterogeneity of blastic crisis. The location of molecular breakpoints within the bcr appeared to be shifted to 5' breakpoint in most of patients with lymphoid blastic crisis, but varied in myeloid blastic crisis. 概要: The cloning of human granulocyte colony-stimulating factor (hG-CSF) has been recently established. Thereafter, purified recombinant hG-CSF have been able to be used for study. However there is no report demonstrating the distribution of G-CSF in human organs. In order to clarify this point, we made ginea pig antisera against hG-CSF, and immunohistological studies were done. A human CSF producing cancer cell line SPT-3.25, established in our laboratory, was also examined as a positive control. The immunohistological study revealed that G-CSF like substance is present in SPT-3.25 cells, chief cells of gastric gland, exocrine cells of pyloric gland, salivary grand and pancreas and syncytial trophoblast of placenta. On the other hand, it was absent in liver, spleen, lung, kidney and lymph node. For the purpose of determining the expression hG-CSF messenger RNA, northern blotting analysis was then done using 40-mer oligo DNA encoding hG-CSF. Poly (A^+) RNA extracted from SPT-3.25 was hibridezed with the probe, and the messenger size appeared to be 1.5kb. However none of poly (A^+) RNA extracted from spleen, gastric mucosa and placenta was found to be hibridized with it. Based on this preliminary study, G-CSF content must be analized in the extract of organs which were positively stained with anti G-CSF antisera, to determine if these organs be able to produce G-CSF in vivo. 続きを見る

概要: In order to apply a simple purging method by heat to autologous bone marrow transplantation (ABMT), we have revaluated t he ability to purge clonogenic leukemic cells from the simulated marrow mixture of normal marrow cells and leukemic cell lines (HL60, Molt-3 and HEL)in vitro by heat, using two different clonogenic assays for normal granulocyte-macrophage progenitors (CFU-GM) and leukemic cell lines. The cytotoxic effect of cryopreservation on heat-treated normal marrow cells in vitro was also examined. It appeared that in vitro hyperthermia (42℃ for 120 min) is able to selectively remove clonogenic leukemic cells from simulated tumor cell-normal marrow mixtures even when leukemic cell concentrations are increased up to 3×10^6 cells/ml in vitro, and results in a 4-to-6 log destruction of clonogenic leukemic cells/ml according to a limiting dilution assay, while leaving half of normal CFU-GM surviving. The hyperthemic purging of clonogenic leukemic cells was not affected in the presence of normal marrow cells in vitro. This high level of clonogenic leukemic cell depletion by heat correlated with that of immunologic and pharmacologic studies. In addition, treatment of normal or remission marrow cells with hyperthermia did not significantly change the growth of CFU-GM after cryopreservation, and the percentage recovery of CFU-GM ranged from 34 to 79%. These results suggest that in vitro hyperthermia could be applied effectively and safely for the elimination of residual clonogenic leukemic cells in autologous marrow grafts before ABMT. 続きを見る

概要: Intensive chemoradiotherapy with autologous bone marrow transplantation (ABMT) has been explored as a treatment for hema tological malignancies. Ten cases with acute leukemia and Hodgkin's disease were treated with high dose chemotherapy followed by autologous marrow cells (2 unpurged, 8 purged). Six leukemia relapse were observed between day 28 to 331 days after ABMT in 9 patients with hematological reconstitution. One case was dead of interstitial pnemonitis and two are alive without relapse. Attempts to remove minimal residual disease from the harvested marrow have been made by several ex vivo purging methods, but the effects of such procedures have not yet been clarified. 概要: Primary intracranial germ cell tumors, malignant lymphomas, and medulloblastomas have been proved to be very sensitive t o chemotherapy (cisplatin-or carboplatin-containing regimens). Recently we tried the high-dose chemotherapy followed by autologous bone marrow transplantation and autologous peripheral blood stem cell transplantation for two patients with relapsed primary brain tumors.
