概要: At least nine inherited neurodegenerative diseases are known to be caused by expanded CAG repeats encoding polyglutamine stretches. Although cytotoxicities of expanded polyglutamine stretches have been suggested, the molecular mechanisms of neurodegeneration remain unclear. We demonstrated that nuclear translocation of mutant proteins containing expanded polyglutamine stretches is a prerequisite for the expression of their cytotoxicity. Hypothesizing that nuclear proteins that interact with mutant proteins, particularly, those that bind to the expanded polyglutamine stretches, are involved in the pathogenetic mechanisms underlying neurodegeneration, we screened nuclear proteins for their ability of binding to expanded polyglutamine stretches. We found that expanded polyglutamine stretches preferentially bind to TAF_II 130, a coactivator involved in CREB-dependent transcriptional activation. The binding of TAF_II 130 with expanded polyglutamine stretches strongly suppress CREB-dependent transcriptional activation, suggesting that interference with transcription due to the binding of expanded polyglutamine stretches with TAF_II 130 and redistribution of TAF_II 130 are involved in the pathogenetic mechanisms underlying neurodegeneration. 続きを見る

概要: It is an unsolved question how the neuron-specific genes are . regulated in the mature neurons. The neuronal regulation of neuropeptide Y (NPY) gene which is a cotransmitter of sympathetic neurons and expressed abundantly in the central nervous system. The regulation of this gene resembles that of tyrosine hydroxylase gene.Both the membrane depolarization induced by neural activity and neurotrophic factors such as NGF induced the transcriptiional activation of NPY and enkephalin genes. These mechanisms are due to both Ca/almodulin and MAPK (mitogen-activated protein kinase) dependent processes, respectively. These findings indicated that the transcriptional activations of neural genes are due to the novel transactivational mechanisms. The novel transcriptional factors will be reported. 続きを見る

概要: Spot 35 protein was first discovered from the cerebellum as a specific protein. Immunohistochemistry demonstrated wide distribution of this protein in the Purkinje cells, intestine, sensory organs and kidney. cDNA cloning and nucleotide sequence showed that Spot 35 protein is identical with calbindin D 28k, a vitamin D dependent calcium binding protein. Two vitamin D response elements (pDRE and dDRE) were found in the 5' upstream region of the Spot 35/Calbindin D 28k gene. Promoter activities were determined by using luciferase as a reporter gene in MDBK cells which produced endogenously Spot 35/Calbindin D 28k. Transcription of the chimeric gene containing the pDRE was enhanced by 1, 25 (OH)_2D_3 stimulation in an orientation-dependent fashion. 続きを見る
