functions of Golgi apparatus in osteoclasts, we observed the ultrastructural and cytochemical changes resulting from the administration of brefeldin A (BFA), an inhibitor of Golgi structures, and wortmannin (WT), an inhibitor of phosphatidylinositol 3-kinase (PI3-kinase). When treated with BFA, osteoclasts showed a dissociated Golgi apparatus, while the peanut agglutinin (PNA)-positive ruffled borders were poorly developed or disappeared. Moreover, the acid phosphatase (ACPase) activity was somewhat reduced. WT-treated osteoclasts, on the other hand, exhibited no ultrastructural alteration of the Golgi apparatus, but were marked by the disappearance of PNApositive ruffled borders, and a consequent increase in the number of ACPase-positive vesicles and vacuoles in the cytoplasm. These results indicate that the Golgi apparatus of osteoclasts not only plays an important role in the formation of lysosomal enzymes but also the supply of ruffled border membranes. Furthermore, PI 3-kinase appears to be involved in the transport of lysosomal enzymes and the maintenance of ruffled borders.

spleen cells and osteoblastic cells in the co-cultures. Light microscopic investigations revealed that tartrate-resistant acid phosphatase (TRAP)positive mononuclear and multinucleated spleen cells appeared in the vicinity of alkaline phosphatase (ALP)-positive osteoblastic cells. Ultrastructurally, spleen cells extended long cytoplasmic filopodia in all directions, by which spleen cells touched adjacent osteoblastic cells. The adjacent osteoblastic cells and spleen cells adhered to each other by forming electron-dense cytoplasmic materials on their inner leaflets of plasma membranes at cell-cell contact sites. Some adjacent osteoblastic and spleen cells made contact on the plasma membranes, forming an extracellular microenvironment. Coated pits were also formed on the plasma membranes facing this microenvironment. These morphological features of the cell-cell contact between osteoblastic cells and spleen cells indicate that there is internalization of organic components, i.e., receptor-mediated endocytosis in the contact sites between the two types of cells.

of transforming growth factor-β (TGF-β), as well as that of the TGF-βreceptor were examined during the chondrification of ligamentous fibroblasts, by inducing ossification in ligamenta flava of murine lumbar spines, employing recombinant human bone morphogenetic protein-2 (rhBMP-2). Normal ligamenta flava, consisting of flattened fibroblasts, showed no immunolocalization of TGF-βisoforms. From the second week following BMP-administration, cartilage gradually replaced the ligaments. Cells in the central portion of the ligament contained abundant Golgi apparatus as well as enlarged endoplasmic reticula. Proliferative or hypertrophic chondrocytes in this area showed immunoreactivity to TGF-β3 latency-associated peptide, while calcified hypertrophic chondrocytes accompanied by both matrix vesicleand collagen-calcification were noticed at the insertion site to spinal lamina, where there was no immunolocalization of this peptide. In addition, the TGF-βtype I receptor was localized on the proliferative and hypertrophic cells of the central portion. Therefore, as regards the pathological chondrification of ligamentous fibroblasts induced by exogenous BMP-2, TGF-βis speculated to promote matrix-formation and chondrocytic maturation, under the autocrine/paracrine system, in concert with exogenous BMP-2.

When β-1,3-glucanase was induced with pachyman, a β-1,3-glucose polymer obtained from the tree fungus Poria cocus Wolf, six distinct active molecules of the enzyme with different molecular weights were detected in the culture supernatant of this bacterium. Molecular cloning of one of the β-1,3-glucanase genes into E. coli was achieved by transforming E. coli HB101 cells with recombinant plasmids composed of chromosomal DNA fragments prepared from B. circulans WL-12 and the plasmid vector pUC 19. A recombinant plasmid containing 4.4kb of inserted DNA in the PstI site of pUC 19, designated as pNT003, conferred the ability to degrade pachyman on E. coli cells. The presence of pNT003 was harmful for E. coli cells and caused cell lysis, especially at higher temperatures of cultivation. β-1,3-Glucanase activity detected in E. coli was mainly recovered in the periplasmic fraction when cell lysis did not occur. SDS-PAGE analysis revealed that the periplasmic fraction contained four active molecules of β-1,3-glucanase which corresponded to four of the six active molecules produced by B. circulans WL-12.

the reactivity of each anion-exchange resin, the OT-for-OH exchange reaction between each anion-exchange resin and HTO water was studied within the range of 20°C to 80°C in the equilibrium state. In addition, the apparent dissociation constant (pK′) for each resin was estimated by the method of pH-titration. From the observation of the OT-for-OH exchange reaction between each of the following anion-exchange resins and HTO water, the next three items have been found: (1) in the case of acryl-type anion-exchange resin, the effect of temperature on OT-for-OH exchange reaction is larger than that on the reaction in the case of styrene-type one; (2) the radioactivity incorporated into weakly basic anion-exchange resin (OH--form) by the isotope exchange reaction is smaller than that into strongly basic one; (3) in the case of the weak-ly basic anion-exchange resin, the amount of radioactivity incorporated into Cl--form resin by the hydrolysis is larger than the amount of radioactivity incorporated into OH--form resin by the isotope exchange reaction.

o measure large rotation angles accurately. Two parallel PIP’s that have different periods are used to measure a rotation angle of an object. The measurement made with a small-period PIP provides a high accuracy, and the measurement made with a large-period PIP provides a wide range. An accurate measurement for wide-range angles is made by combining the two measured values. The accuracy of the phase detection is determined by the periods of two PIP’s. Rotation angles from approximately −30 to 30 arc min can be measured with an accuracy of 0.2 arc sec. Analytical results are supported by experimental results.

asuring two-dimensional (2D) small rotation angles by using two different PIP's that are orthogonal to each other is proposed. We simultaneously measure the 2D small rotation angles Δθ and ΔΦ by detecting the phases of the orthogonal PIP's reflected by an object at two detection points. A sensitivity of4.9 mrad/arcsec and a spatial resolution of 1.5 × 1.5 mm^2 are achieved in the measurement. Theoretical analysis and experimental results show that errorε1 in the measurement of ΔΦ is almost equal to -0.01Δθ and errore ε2 in the measuremcnt of Δθis almost equal to -0.01A ΔΦ. For small rotation angles of less than a few tens of arcseconds, the random errors whose standard deviations are 0.6 arcsec are dominant.

A high sensitivity, or a high ratio of the phase change to the rotation angle, 17 mrad/arcsec, can be achieved by determining the positions of two detection points. A high spatial resolution of ~0.5 mm is also obtained. We analyze the measurement error caused by the alignment of the parallel interference pattern and a random measurement error caused by the phase detection. The theoretical analyses and the experimental results make the characteristics of the method clear and show that the method has an accuracy of 0.2 arcsec for small rotation angles.

A novel method was developed, that includes the measurement of the film thickness profile on micron-sized trenches and the molecular diffusivity of the growth species. The molecular size was about 1 nm at 700-850℃ and decreased gradually with increasing temperature. The sticking probability increased from 0.02 to 0.5 in the temperature range 700-950℃ and, surprisingly, obeyed the Arrhenius law in spite of this large probability of sticking. The activation energy amounted to 136 kJ/mol.

are responsible for the stacking structure with glutamine, into aliphatic amino acids gave a significantly lower temperature for the catalytic optimum as for the mutageneses of QW motifs 4, 5a and 5b, which are specifically present in SHCs. However, there was no change in the optimal temperatures of the mutated SHCs targeted at the other five motifs 1, 2, 3, 5c and 6. Thus, reinforcement against heat denaturation can be proposed as a function of the three QW motifs 4, 5a and 5b, but no function could be identified for the QW motifs 1, 2, 3, 5c and 6, although they are commonly found in all the families of prokaryotic SHCs and eukaryotic oxidosqualene cyclases. On the other hand, the three conserved tryptophans of W169, W312 and W489, which are located inside the putative central cavity and outside the QW motifs, were identified as components of the active sites, but also had a function against thermal denaturation. The other two tryptophan residues of W142 and W558, which are located outside the QW motifs, were found not to be active sites, but also had a role for stabilizing the protein structure. It is noteworthy that the mutants replaced by phenylalanine had higher temperatures for the catalytic optimum than those replaced by aliphatic amino acids. The catalytic optimal pH values for all the mutants remained unchanged with an identical value of 6.0.

assembly was positioned anally into the ileal W pouch. We distended the ileal W pouch with the balloons by 2 mmHg increments for 2 min. applied every 1 min. and compliance of the pouch was measured. Patients declared the threshold of distending perception, threshold sense of defecation and defecating urgency. Furthermore, patients answered our formatted questions about postoperative outcome of functional scores. We concluded that 1 ) the compliance when patients declared threshold sense of defecation correlated well with the maximally tolerable volume for defecation, and it reflected the capacity of reserving stool, 2 ) the function of the ileal pouch has improved with postoperative period, and 3) the improvement of subjective functional scores were associated with compliance increase of the ileal pouch and decrease of PVEs /hour (phasic volume events/hour).

tty acid metabolism and myocardial viability. Some investigators reported absence of BMIPP-accumulation in the hearts of few patients with heart diseases. The mechanism and clinical significance of absence of BMIPP-accumulation in heart are not known. CD36, glycoprotein IV expressed in platelet membrane, is a receptor for collagen, thrombospondin, oxidised LDL and long-chain fatty acids. CD36 may play a role in platelet aggregation as well as in formation and progression of arteriosclerosis, and in myocardial fatty acid metabolism. The expression and genetic abnormality of CD36 were investigated in this study in relation to myocardial accumulation of BMIPP. There are two types of platelet CD36 deficiency; types I and II , associated with the absence and presence of CD36 on monocytes, respectively. In 200 patients with various heart diseases in this study, 24 cases revealed CD36 deficiency including 8 type I and 16 type II. No myocardial BMIPP-accumulations were observed in all 8 type I cases, while significant accumulation was seen in all type II cases. Genetic analysis of CD36 was carried in 3 of 8 cases with type I deficiency. Deletions of 52 base inn exon 4 and 70 base in exon 9 were documented in mRNA of CD36 in all 3 cases by means of sequence analysis after RT-PCR. Genomic DNA of CD36 was also analyzed by using a PCR-direct sequence method, and there were two types of point mutation, namely, ^<970>T→C/Ile-Thr (one of 3 cases) and ^<478>C→T/Pro→Ser (other 2 cases). The point mutation at ^<478>T was previously reported by other researchers. This is the first report to show the alternative splicing in exons 4 and 9 and the point mutation at ^<970> C. CD36 deficiency may contribute the pathogenesis of heart diseases and arteriosclerosis through abnormal fatty acid metabolism.

concentrations of the undissociated form, the antimicrobial activities of acetic and propionic acids were higher than that of lactic acid, irrespective of test microorganisms. In a single cultivation of Bifidobacterium longum, a mixture of lactic (17g/l) and acetic (20g/l) acids was produced from 50g/l lactose and its antimicrobial activities against M. luteus, Pseudomonas sp., and S. aureus correspond to that of 32, 19, and 25g/l of acetic acid, respectively. To increase the total antimicrobial activity, a co-culture of B. longum and Propionibacterium freudenreichii, in which lactic acid produced once from lactose by B. longum was converted to acetic and propionic acids by P. freudenreichii, was done using TPY medium containing commercially available peptones as a nitrogen source. By the sequential conversion of lactose using the two microorganisms, the culture supernatant containing a mixture of acetic (27g/l) and propionic (13g/l) acids without lactic acid was produced. The antimicrobial activities of the mixture against M. luteus, Pseudomonas sp., and S. aureus were 35, 30, and 26g/l as a concentration of acetic acid, respectively, higher than that obtained in the cultivation of B. longum alone. When the medium containing an enzymatic hydrolyzate of whey proteins with a protease was used in the co-culture of B. longum and P. freudenreichii, the culture supernatant containing the mixture of organic acids was also obtained in the same manner as the co-culture using TPY medium and the activities were 43, 29, and 29g/l as a concentration of acetic acid for M. luteus, Pseudomonas sp. and S. aureus, respectively.

2170 is grown in the presence of chitin. The gene (cbp) for CBP21 was found to be located in a region 1.5 kb downstream of the chiB gene encoding chitinase B. The cbp gene encodes a polypeptide of 197 amino acids with a calculated size of 21.6 kDa containing a putative signal sequence of 27 amino acids. Comparison of the amino acid sequence of the deduced polypeptide with that of other proteins showed that CBP21 is similar (45.3% amino acid identity) to CHB1 of Streptomyces olivaceoviridis. Purified CBP21 prepared from the periplasmic fraction of Escherichia coli carrying the cloned cbp gene showed its highest binding activity to squid chitin (β-chitin) followed by colloidal chitin and regenerated chitin. Binding of CBP21 to regenerated chitin was affected by pH, in particular, low pH reduced binding activity markedly. The presence of similar chitin binding proteins in the distantly related microorganisms, Streptomyces and Serratia, suggests a wide distribution of this type of chitin binding protein in chitinolytic microorganisms.
