概要: Osteoclasts secrete protons and lysosomal enzymes through ruffled borders, resulting in demineralization and degradation of bone matrix. Golgi apparatus is thought to play a key role in this process. In order to elucidate the specific functions of Golgi apparatus in osteoclasts, we observed the ultrastructural and cytochemical changes resulting from the administration of brefeldin A (BFA), an inhibitor of Golgi structures, and wortmannin (WT), an inhibitor of phosphatidylinositol 3-kinase (PI3-kinase). When treated with BFA, osteoclasts showed a dissociated Golgi apparatus, while the peanut agglutinin (PNA)-positive ruffled borders were poorly developed or disappeared. Moreover, the acid phosphatase (ACPase) activity was somewhat reduced. WT-treated osteoclasts, on the other hand, exhibited no ultrastructural alteration of the Golgi apparatus, but were marked by the disappearance of PNApositive ruffled borders, and a consequent increase in the number of ACPase-positive vesicles and vacuoles in the cytoplasm. These results indicate that the Golgi apparatus of osteoclasts not only plays an important role in the formation of lysosomal enzymes but also the supply of ruffled border membranes. Furthermore, PI 3-kinase appears to be involved in the transport of lysosomal enzymes and the maintenance of ruffled borders. 続きを見る

概要: We have previously established a mouse co-culture system of osteoblastic cells and spleen cells for examining osteoclast differentiation. In the present study, we examined the morphological features of the cell-cell contact between mouse spleen cells and osteoblastic cells in the co-cultures. Light microscopic investigations revealed that tartrate-resistant acid phosphatase (TRAP)positive mononuclear and multinucleated spleen cells appeared in the vicinity of alkaline phosphatase (ALP)-positive osteoblastic cells. Ultrastructurally, spleen cells extended long cytoplasmic filopodia in all directions, by which spleen cells touched adjacent osteoblastic cells. The adjacent osteoblastic cells and spleen cells adhered to each other by forming electron-dense cytoplasmic materials on their inner leaflets of plasma membranes at cell-cell contact sites. Some adjacent osteoblastic and spleen cells made contact on the plasma membranes, forming an extracellular microenvironment. Coated pits were also formed on the plasma membranes facing this microenvironment. These morphological features of the cell-cell contact between osteoblastic cells and spleen cells indicate that there is internalization of organic components, i.e., receptor-mediated endocytosis in the contact sites between the two types of cells. 続きを見る

概要: In order to elucidate the causes of ossification in spinal ligaments, ultrastructural alteration and immunolocalization of transforming growth factor-β (TGF-β), as well as that of the TGF-βreceptor were examined during the chondrification of ligamentous fibroblasts, by inducing ossification in ligamenta flava of murine lumbar spines, employing recombinant human bone morphogenetic protein-2 (rhBMP-2). Normal ligamenta flava, consisting of flattened fibroblasts, showed no immunolocalization of TGF-βisoforms. From the second week following BMP-administration, cartilage gradually replaced the ligaments. Cells in the central portion of the ligament contained abundant Golgi apparatus as well as enlarged endoplasmic reticula. Proliferative or hypertrophic chondrocytes in this area showed immunoreactivity to TGF-β3 latency-associated peptide, while calcified hypertrophic chondrocytes accompanied by both matrix vesicleand collagen-calcification were noticed at the insertion site to spinal lamina, where there was no immunolocalization of this peptide. In addition, the TGF-βtype I receptor was localized on the proliferative and hypertrophic cells of the central portion. Therefore, as regards the pathological chondrification of ligamentous fibroblasts induced by exogenous BMP-2, TGF-βis speculated to promote matrix-formation and chondrocytic maturation, under the autocrine/paracrine system, in concert with exogenous BMP-2. 続きを見る

概要: Bacillus circulans WL-12, a yeast and fungal cell wall lytic bacterium, secretes a variety of polysaccharide degrading e nzymes into the culture medium. When β-1,3-glucanase was induced with pachyman, a β-1,3-glucose polymer obtained from the tree fungus Poria cocus Wolf, six distinct active molecules of the enzyme with different molecular weights were detected in the culture supernatant of this bacterium. Molecular cloning of one of the β-1,3-glucanase genes into E. coli was achieved by transforming E. coli HB101 cells with recombinant plasmids composed of chromosomal DNA fragments prepared from B. circulans WL-12 and the plasmid vector pUC 19. A recombinant plasmid containing 4.4kb of inserted DNA in the PstI site of pUC 19, designated as pNT003, conferred the ability to degrade pachyman on E. coli cells. The presence of pNT003 was harmful for E. coli cells and caused cell lysis, especially at higher temperatures of cultivation. β-1,3-Glucanase activity detected in E. coli was mainly recovered in the periplasmic fraction when cell lysis did not occur. SDS-PAGE analysis revealed that the periplasmic fraction contained four active molecules of β-1,3-glucanase which corresponded to four of the six active molecules produced by B. circulans WL-12. 続きを見る

概要: In order to reveal the effect of temperature on (i) the dissociation equilibrium of tritiated water (HTO water) and (ii) the reactivity of each anion-exchange resin, the OT-for-OH exchange reaction between each anion-exchange resin and HTO water was studied within the range of 20°C to 80°C in the equilibrium state. In addition, the apparent dissociation constant (pK′) for each resin was estimated by the method of pH-titration. From the observation of the OT-for-OH exchange reaction between each of the following anion-exchange resins and HTO water, the next three items have been found: (1) in the case of acryl-type anion-exchange resin, the effect of temperature on OT-for-OH exchange reaction is larger than that on the reaction in the case of styrene-type one; (2) the radioactivity incorporated into weakly basic anion-exchange resin (OH--form) by the isotope exchange reaction is smaller than that into strongly basic one; (3) in the case of the weak-ly basic anion-exchange resin, the amount of radioactivity incorporated into Cl--form resin by the hydrolysis is larger than the amount of radioactivity incorporated into OH--form resin by the isotope exchange reaction. 続きを見る

概要: A laser-diode (LD) interferometer that uses an accurate photothermal-modulating technique is proposed. Since this technique with the photothermal method modulates only a wavelength of the LD, measurement accuracy is not affected by an intensity modulation that usually appears in the current modulation. The fundamental characteristics of this technique are investigated in detail. The new setup is tested, and its accuracy is compared with that of a previous system. 続きを見る

概要: We propose an interferometer in which the relationship between the degree of coherence (DCH) and the optical path difference (OPD) is utilized for determining an OPD longer than a wavelength. A superluminescent diode is employed as the source of the interferometer, and sinusoidal phase-modulating interferometry is used to detect the DCH and the phase of the interference signal. The combination of the OPD determined from the DCH and the phase of an interference signal enables us to measure an OPD longer than a wavelength with a high accuracy of a few nanometers. Experimental results show clearly the usefulness of the interferometer for a step-profile measurement. 続きを見る

概要: Based on the idea of measuring small rotation angles with a parallel interference pattern (PIP), a method is developed t o measure large rotation angles accurately. Two parallel PIP’s that have different periods are used to measure a rotation angle of an object. The measurement made with a small-period PIP provides a high accuracy, and the measurement made with a large-period PIP provides a wide range. An accurate measurement for wide-range angles is made by combining the two measured values. The accuracy of the phase detection is determined by the periods of two PIP’s. Rotation angles from approximately −30 to 30 arc min can be measured with an accuracy of 0.2 arc sec. Analytical results are supported by experimental results. 続きを見る

概要: We propose a new signal-processing method for eliminating measurement errors that occur in the wavelength-multiplexed ph ase-locked laser diode interferometer. The basic idea proposed here is a very simple but effective way to improve measurement accuracy. With our scheme, the phase in the interference signal is strictly shifted by 2π, which enables us to eliminate measurement errors. The equivalent wavelength Λ is 80 mm, and the measurement accuracy reaches ~Λ/600. A step-height measurement was also carried out in the experiment. 続きを見る
